Glibenclamide stimulates insulin secretion from pancreatic β-cells by binding to the sulfonylurea receptor (SUR1) and inhibiting ATP-sensitive potassium channels, leading to depolarization and insulin release. Metformin reduces hepatic gluconeogenesis and improves peripheral insulin sensitivity through activation of AMP-activated protein kinase (AMPK). Pioglitazone acts as a peroxisome proliferator-activated receptor gamma (PPAR-γ) agonist, enhancing insulin sensitivity in adipose tissue, skeletal muscle, and liver.
