Methotrexate is a folate antagonist that inhibits the enzyme dihydrofolate reductase (DHFR), reducing the production of tetrahydrofolate required for DNA, RNA, and protein synthesis. At lower doses, it also increases extracellular adenosine concentrations, producing anti-inflammatory and immunosuppressive effects that help control autoimmune diseases while inhibiting abnormal cell proliferation.
