Introduction :

Sitagliptin 100mg is an oral diabetes medicine that helps control blood sugar levels. Sitagliptin 100mg works by regulating the levels of insulin your body produces after eating.Sitagliptin is used together with diet and exercise to improve blood sugar control in adults with type 2 diabetes mellitus. Sitagliptin 100mg reduces blood sugar levels by increasing the amount of insulin made and released from pancreas, and also decreases the amount sugar made by liver. Sitagliptin 100mg is a DPP-4 inhibitor (dipeptidyl peptidase-4 inibitor).

Indication :

Sitagliptin 100mg is used alone or together with other medicines (eg, insulin, glimepiride, metformin, or pioglitazone) and with proper diet and exercise to treat high blood sugar levels caused by type 2 diabetes. Sitagliptin 100mg helps to control blood sugar levels by increasing substances in the body that make the pancreas release more insulin. It also signals the liver to stop producing sugar (glucose) when there is too much sugar in the blood. This medicine does not help patients who have insulin-dependent or type 1 diabetes. This medicine is available only with your doctors prescription.

Mechanism of Action :

Sitagliptin is a dipeptidyl peptidase-4 (DPP-4) enzyme inhibitor. The activity of glucagon-like peptide 1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) is limited by the enzyme DPP-4, which rapidly hydrolyzes incretin hormones to form inactive products. Sitagliptin prevents the hydrolysis of incretin hormones by DPP-4, thereby increasing plasma concentrations of the active forms of GLP-1 and GIP. By stimulating the levels of active incretins, sitagliptin increases insulin release and reduces glucagon levels with glucose-dependent behavior. The glucose-dependent sitagliptin mechanism is different from the sulfonylureas mechanism, which increases insulin secretion even when glucose levels are low and can lead to hypoglycemia in patients with type 2 diabetes and normal patients.
